Our peptide reference materials are produced by Fmoc-based solid-phase peptide synthesis (SPPS), followed by reverse-phase HPLC purification and lyophilization under controlled-environment conditions. All synthesis is performed by contracted ISO-aligned manufacturing partners in the United States and Germany.
Process overview
- Resin loading — Wang or Rink-Amide resin selected for the target C-terminus.
- Iterative coupling — HBTU/HOBt or Oxyma/DIC activation, Fmoc deprotection with 20% piperidine in DMF.
- Global deprotection & cleavage — TFA cocktail (TFA/TIS/H₂O/EDT) with appropriate scavengers.
- Precipitation — cold diethyl ether, centrifugation, wash cycles.
- Purification — preparative RP-HPLC on C18, ACN/H₂O + 0.1% TFA gradient.
- Lyophilization — primary & secondary drying to a stable white powder.
- Analytical release — HPLC purity, MS identity, water content, appearance.
